Indications
- Neuropathic pain associated with diabetic peripheral neuropathy (DPN)
- Postherpetic neuralgia (PHN)
- Adjunctive therapy for the treatment of partial-onset seizures in patients 1 month of age and older
- Fibromyalgia
- Neuropathic pain associated with spinal cord injury
Pharmacology
Pregabalin is a structural derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA). It binds with high affinity to the alpha2-delta site in central nervous system tissues.
Dosage
- Neuropathic pain associated with diabetic peripheral neuropathy in adults (DPN): 50-300 mg/day
- Postherpetic neuralgia in adults (PHN): 75-300 mg/day
- Management of fibromyalgia in adults: 300-450 mg/day
- Neuropathic pain associated with spinal cord injury in adults: 150-600 mg/day
Administration
- Route of administration: Oral
- Special administration for Pregabalin CR tablet: Administer after an evening meal, swallow whole
Interaction
- Drug interaction with medication: Unlikely to cause significant pharmacokinetic drug interactions
Contraindications
Contraindicated in patients with known hypersensitivity to Pregabalin or any of its components
Side Effects
- Dizziness
- Somnolence
- Dry mouth
- Edema
- Blurred vision
- Weight gain
- Difficulty with concentration/attention (primarily)
Pregnancy & Lactation
- Pregnancy: No adequate and well-controlled studies
- Lactation: Small amounts detected in the milk of lactating women, not recommended during treatment
Precautions & Warnings
- Angioedema can occur and may be associated with life-threatening respiratory compromise
- Discontinue immediately in case of hypersensitivity reactions
- Increased risk of suicidal thoughts or behavior
- May cause dizziness and somnolence, impairing ability to drive or operate machinery
- May cause peripheral edema, use caution when coadministering with thiazolidinedione antidiabetic agents
Use in Special Populations
Safety and effectiveness not established in pediatric patients for certain indications
Overdose Effects
Reduced consciousness, depression/anxiety, seizures, heart block; no specific antidote
