Indications
- Neuropathic pain associated with diabetic peripheral neuropathy (DPN)
- Postherpetic neuralgia (PHN)
- Adjunctive therapy for the treatment of partial-onset seizures in patients 1 month of age and older
- Fibromyalgia
- Neuropathic pain associated with spinal cord injury
Pharmacology
Structural derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA)
Dosage
- Maximum recommended dose of Pregabalin for DPN: 300 mg/day
- Maximum recommended dose of Pregabalin for PHN: 300 mg/day
- Maximum recommended dose of Pregabalin for fibromyalgia: 450 mg/day
- Maximum recommended dose of Pregabalin for spinal cord injury: 600 mg/day

Administration
Oral route, can be taken with or without food, Pregabalin CR tablet should be administered after an evening meal, gradual tapering when discontinuing
Interaction
Unlikely to be involved in significant pharmacokinetic drug interactions, no specific drug interaction with food
Contraindications
Known hypersensitivity to Pregabalin or any of its components

- Somnolence
- Dry mouth
- Edema
- Blurred vision
- Weight gain
- Thinking abnormalities
- Increased weight and increased appetite in pediatric patients
Pregnancy & Lactation
No adequate and well-controlled studies in pregnant women, small amounts detected in the milk of lactating women, breastfeeding not recommended
Precautions & Warnings
- Angioedema
- Hypersensitivity reactions
- Increased risk of suicidal thoughts or behavior
- Respiratory depression
- Dizziness and somnolence
- Peripheral edema
- Cautious co-administration with thiazolidinedione antidiabetic agents
Use in Special Populations
- Safety and effectiveness not established in pediatric patients
- Safety and effectiveness not established for pediatric patients below the age of 1 month
- Safety and effectiveness of Xablin extended-release tablet not established in pediatric patients
Overdose Effects
Reduced consciousness, depression/anxiety, confusional state, agitation, restlessness, seizures, heart block
