Dosage

  • Maximum recommended dose of 100 mg three times a day
  • Dosing should begin at 50 mg three times a day
  • Recommended doses of 75 to 150 mg two times a day
  • Recommended dose of 300 to 450 mg/day for fibromyalgia
  • Recommended dose range of 150 to 600 mg/day for spinal cord injury

Route of Administration

  • Oral route
  • Can be taken with or without food
  • Pregabalin CR tablet should be administered after an evening meal
  • Should be swallowed whole and not split, crushed, or chewed

Pharmacology

  • Structural derivative of GABA
  • Binds with high affinity to alpha2-delta site in central nervous system tissues

Indications

  • Neuropathic pain associated with diabetic peripheral neuropathy (DPN)
  • Postherpetic neuralgia (PHN)
  • Adjunctive therapy for the treatment of partial-onset seizures
  • Fibromyalgia
  • Neuropathic pain associated with spinal cord injury

Contraindications

  • Known hypersensitivity to Pregabalin or its components

Overdose Effects

  • Signs and symptoms include reduced consciousness, depression/anxiety, confusional state, agitation, restlessness, seizures, and heart block
  • No specific antidote
