Indications
- Treatment of all types of epilepsy

Composition
- Sodium Valproate tablet 200 mg
- Sodium Valproate syrup 200 mg/5ml
- Sodium Valproate controlled release 200 tablet

- Sodium Valproate injection 500 mg/5ml
Pharmacology
- Endowed with anti-epileptic activity
- Mechanism related to increased brain levels of gamma-aminobutyric acid (GABA)
Dosage & Administration
- Oral administration
- Initial dosage for epilepsy: Adults - 600 mg daily, Children (over 20 kg) - 400 mg daily, Children (up to 20 kg) - 20 mg/kg daily
- Maintenance dosage for epilepsy: 1-2 g daily for adults, 20-30 mg/kg daily for children
- Dosage for bipolar disorder: Initially 600 mg daily, Maintenance dosage 1000-2000 mg daily
- Dosage for prophylaxis of migraine: 400-600 mg daily, may require 1000-1500 mg daily
- Injection administration: IV injection over 3-5 minutes, continuous or repeated infusion
Interaction
- Non-specific inhibitor of drug metabolism
- Significant interaction with Phenobarbital, Phenytoin, Warfarin, Aspirin
Contraindications
- Known hypersensitivity to the drug
- Liver dysfunction
- Restricted use during pregnancy and in women of childbearing potential
Side Effects
- Anorexia, nausea, vomiting
- CNS effects: sedation, ataxia, tremor
- Minimized side effects with enteric coated tablets
- Rash, alopecia, stimulation of appetite
- Elevation of liver enzymes in plasma
- Hepatic injury, acute pancreatitis, hyperammonemia
- Fulminate hepatitis (rare)
Pregnancy & Lactation
- Crosses the placenta
- Associated with neural tube defects in first trimester exposure
- Excreted in breast milk, probably causes no risk to the child
Precautions & Warnings
- Monitoring of liver functions
- Ensuring no undue potential for bleeding
- Care in renal impairment, pregnancy, breast-feeding, systemic lupus erythematosus
- False interpretation of urine ketone test due to metabolite
- Avoid sudden withdrawal of therapy
