Indications

  • Treatment of all types of epilepsy
  • Prophylaxis of febrile convulsion
  • Prophylaxis of post-traumatic epilepsy

Composition

  • Sodium Valproate tablet - 200 mg
  • Sodium Valproate syrup - 200 mg/5 ml
  • Sodium Valproate controlled release 200 tablet - 200 mg
  • Sodium Valproate controlled release 300 tablet - 300 mg
  • Sodium Valproate controlled release 500 tablet - 500 mg
  • Sodium Valproate injection - 500 mg/5 ml

Pharmacology

  • Anti-epileptic activity against a variety of seizures
  • Mechanism may be related to increased brain levels of gamma-aminobutyric acid (GABA)

Dosage & Administration

  • Oral and injection administration
  • Dosage varies for epilepsy, bipolar disorder, prophylaxis of migraine
  • Dosage adjustments for adults and children

Interaction

  • Inhibitor of drug metabolism
  • Significant interaction with Phenobarbital, Phenytoin, Warfarin, Aspirin

Contraindications

  • Hypersensitivity to the drug
  • Liver dysfunction
  • Restricted use during pregnancy and in women of childbearing potential

Side Effects

  • Anorexia, nausea, vomiting
  • Effects minimized with enteric-coated tablets
  • CNS effects include sedation, ataxia, tremor
  • Rarely, fulminate hepatitis may develop
  • Elevation of liver enzymes in plasma observed in up to 40% of patients

Pregnancy & Lactation

  • Crosses the placenta
  • Associated with neural tube defects in newborn
  • Excreted in breast milk

Precautions & Warnings

  • Monitoring of liver functions
  • No undue potential for bleeding before major surgery
  • Care in renal impairment, pregnancy, breast-feeding, systemic lupus erythematosus
  • Partial elimination in the urine as a ketone metabolite
