Indications

Description
- Levodopa as metabolic precursor of dopamine
- Carbidopa as dopa decarboxylase inhibitor
- Conversion of levodopa to dopamine in brain
Dosage & Administration
- Initial dosage with 100/10 mg tablet
- Titration upward for optimum carbidopa dosage
- Starting dosage with 250/25 mg tablet
- Individualized therapy for desired response
- Maintenance dosage for levodopa requirement
- Limited experience with high carbidopa dosage
Interaction
- Symptomatic postural hypotension with antihypertensive medicines
- Rare adverse reactions with tricyclic antidepressants
- Decrease in bioavailability with ferrous sulphate or gluconate
- Reduction of therapeutic effects with dopamine-2 receptor antagonists
- Reversal of effects by phenytoin and papaverine
- Severe orthostatic hypotension with selegiline
Contraindications
- Hypersensitivity to carbidopa and levodopa
- Narrow-angle glaucoma
- Risk of malignant melanoma activation
Side Effects
- Central neuropharmacologic adverse effects
- Diminishable reactions with dosage reduction
- Common adverse effects such as dyskinesias and nausea
- Various adverse effects across body systems
Pregnancy & Lactation
- Unknown effects on human pregnancy

Precautions & Warnings
- Not recommended for medicine-induced extrapyramidal reactions
- Careful observation for depression with suicidal tendencies
- Caution in patients with severe cardiovascular or pulmonary disease
- Monitoring of cardiac function in patients with arrhythmia
- Cautious treatment of patients with wide-angle glaucoma
