Indications
- Adjunctive treatment of all forms of parkinsonian syndrome
- Symptomatic treatment of idiopathic, postencephalitic, and arteriosclerotic parkinsonian
- Control of extrapyramidal symptoms induced by neuroleptic drugs
Pharmacology
- Antimuscarinic antiparkinsonian agent
- Corrects relative cholinergic excess in parkinsonian
- Rapid absorption from gastrointestinal tract
- Duration of effect up to 4 hours
Dosage & Administration
- Oral administration after meals
- Initial dosage for parkinsonism: 2.5 mg 3 times a day
- Initial dosage for drug-induced extrapyramidal symptom: 2.5 mg 3 times a day
- Maximum daily IM or IV injection: 20 mg
Interaction
- Increased anticholinergic activity with agents having anticholinergic amantadine
- Reduced absorption of ketoconazole by concomitant administration of procyclidine
Contraindications
- Caution in children and geriatric patients
- Cautious use in patients with diarrhea, cardiovascular disease, glaucoma, urinary retention, hepatic or renal impairment
- Unestablished safety during pregnancy and lactation
Side Effects
- Dryness of the mouth at usual dosage levels
- Occasional adverse effects: mydriasis, blurred vision, G.I. effects, allergic reaction, muscular weakness
- High doses may cause vertigo, confusion, and hallucination
Pregnancy & Lactation
- Safe use requires weighing potential benefits against possible hazards to the mother and child
Precautions & Warnings
- Precaution needed in case of hepatic and renal impairment
- Caution in children, elderly, pregnancy, and lactation
- Avoid abrupt withdrawal to prevent rebound Parkinsonism symptoms
