Indications
- Treatment of obese patients with BMI > 30 kg/m^2
- Treatment of overweight patients with BMI > 28 kg/m^2
- Treatment of patients with associated risk factors like type II diabetes and hyperlipidemia
- Treatment of obese adolescents
- Adequate reduction of body weight
- Treatment with mildly hypocaloric diet
Pharmacology
- Specific and long-acting lipase inhibitor
- Forms covalent bond with active serine site of gastric and pancreatic lipases
- Inactivated enzyme rendering unable to hydrolyze dietary fats
- Reduction in cyclosporine plasma levels
Dosage & Administration
- Recommended dose of 120 mg capsule
- To be taken immediately before, during or up to one hour after each main meal
- Increase in fecal fat 24-48 hours after dosing
- Dose omission if meal contains no fat
Interaction
- Inhibition of absorption of vitamin E acetate supplement
- Possible effect on absorption of vitamin D, vitamin A and nutritionally-derived vitamin K
- Reported hypothyroidism when coadministered with levothyroxine
- Potential decrease in vitamin K absorption
Contraindications
- Chronic malabsorption syndrome
- Cholestasis
- Hypersensitivity to orlistat or other ingredients of the capsules
