- Symptoms Treated:
- Mild to Moderately Severe Alzheimer's Dementia
- Mild to Moderately Severe Dementia in Idiopathic Parkinson's Disease
- Pharmacology: Carbamate derivative related to physostigmine, cholinesterase inactivation, brain acetylcholinesterase specificity
- Dosage Form:
- Capsule
- Transdermal Patch
- Dosage Instructions:
- Capsule:
- Initial Dose: 1.5 mg twice a day
- Dose Titration: 1.5 mg to 6 mg twice a day based on tolerability
- Maintenance Dose: 3 mg to 6 mg twice a day
- Re-initiation of Therapy: Start at 1.5 mg twice daily
- Transdermal Patch:
- Initial Dose: 4.6 mg/24 hours
- Dose Titration: 4.6 mg to 13.3 mg/24 hours based on tolerability
- Specific Disease Doses:
- Mild to Moderate Alzheimer's & Parkinson's Disease Dementia: 9.5 mg/24 hours or 13.3 mg/24 hours once daily
- Severe Alzheimer's Disease: 13.3 mg/24 hours once daily
- Treatment Interruption: Retitrate dosage starting at 4.6 mg per 24 hours
- Capsule:
- Administration: Twice a day, with morning and evening meals
- Interactions:
- Anaesthesia: Exaggerated effects on succinylcholine-type muscle relaxants
- Other Medications:
- Cholinomimetic Substances
- Anticholinergic Medicinal Products
- Pharmacokinetic Interaction: None observed with digoxin, warfarin, diazepam, or fluoxetine
- Cardiac Conduction: No untoward effects observed with digoxin
- Contraindications: Hypersensitivity to the active substance or other carbamate derivatives
- Side Effects: Most commonly gastrointestinal, including nausea and vomiting
