Indications

  • Reduces blood sugar levels
  • Used in type 2 diabetes Mellitus in adults
  • When diet, exercise, and weight loss alone do not control blood sugar

Pharmacology

  • Stimulates insulin release from pancreatic cells
  • Facilitates Ca+2 transport across cell membranes
  • Decreases hepatic glucose output

Dosage & Administration

  • Usual initial dose: 40 to 80 mg daily
  • Maximum dose: 320 mg daily in divided doses
  • Should be taken before meal
  • Modified release preparation: recommended daily dose is 1 to 4 tablets (max 120 mg) at breakfast

Interaction

  • May interact with oral antidiabetics, antibiotics, beta-blockers, ACE-inhibitors, fluconazole, corticosteroids, St John's Wort, and more
  • May increase effects of warfarin
  • Not recommended with alcohol consumption

Contraindications

  • Allergy to sulfonylurea group or related medicines
  • Insulin-dependent diabetes (type 1)
  • Severe kidney or liver disease
  • Breastfeeding
  • Taking medicines for fungal infections
