Amantril 100 mg (Capsule)

Indications

• Treatment of parkinsonism
• Treatment of drug-induced extrapyramidal reactions
• Prophylaxis and treatment of signs and symptoms of infection caused by various strains of influenza A virus

Pharmacology

• Mechanism of antiviral activity
• Inhibition of influenza A virus replication
• Variability of in vitro susceptibility to amantadine

Dosage & Administration

• Parkinson's disease:
  • Initial adult dose of 100mg/day
  • Max dose of 400mg daily
• Prophylaxis of influenza A:
  • Adult dose of 100mg daily for up to 6 weeks
  • Child dose of 100mg daily for 10-15 year olds
• Influenza A:
  • Adult dose of 100mg daily for 5 days
  • Elderly dose of 1 day
• Herpes zoster in immunocompromised patients:
  • Adult dose of 100mg bid for 14 days, continued for another 14 days if pain persists

Interaction

• Potential interaction with anticholinergic agents
• Additive CNS toxicity with concurrent use of certain drugs or substances

Contraindications

• Known hypersensitivity to the active substances or excipients

• Edema of ankles
• Anxiety
• Hallucinations
• Dry mouth
• Anorexia
• Palpitations
• Constipation
• Diaphoresis

Pregnancy & Lactation

• Pregnancy category C
• Excretion into breast milk in low concentrations

Precautions & Warnings

• Avoid abrupt discontinuation in Parkinson's disease patients
• Monitor for atropine-like effects when used concurrently with other drugs
• Potential for precipitating delirium and other psychiatric reactions upon abrupt discontinuation

Use in Special Populations

• Patients with renal impairment:
  • Different dosage based on creatinine clearance
• Patients with hepatic impairment:
  • Use with caution
