Indications
- Adjunct to diet and exercise for Type II diabetes mellitus
- Can be used in combination with metformin and insulin
- Helps lower blood glucose levels
Pharmacology
- Sulfonylurea antidiabetic agent
- Stimulates insulin release from pancreatic beta cells
- Improves sensitivity of beta cells to glucose stimulus
Dosage
- Initial dose of 1 mg once daily
- Gradual dose titration based on glucose monitoring
- Usual dose range of 1 to 4 mg daily
Administration
- Tablet must be swallowed with sufficient liquid
Interaction
- Interactions with ACE inhibitors, aminosalicylic acid, anabolic steroids, etc.
- Drugs that may potentiate or attenuate hypoglycaemic action
Contraindications
- Not suitable for Type I diabetes mellitus, diabetic ketoacidosis, or coma
- Contraindicated in patients hypersensitive to Glimepiride
- Should not be used in severe hepatic or renal dysfunction
Side Effects
- Hypoglycaemia
- Temporary visual impairment
- Nausea, vomiting, diarrhoea
- Abdominal pain
- Urticaria
- Fall in blood pressure
Pregnancy & Lactation
- Not recommended during pregnancy, changeover to insulin necessary
- Should not be taken while breastfeeding
Precautions & Warnings
- Careful monitoring for hypoglycaemia in initial weeks of treatment
- Immediate intake of carbohydrates to control hypoglycaemia
Overdose Effects
- Overdosage may produce hypoglycaemia
- Mild symptoms can be treated with oral glucose
- Severe reactions require immediate hospitalization
