Indications
- Lower blood glucose in noninsulin dependent diabetes mellitus (NIDDM) patients
- Adjunct to diet and exercise
- May be used concomitantly with metformin
- Use in combination with insulin to lower blood glucose
- Potential for hypoglycemia with combined use of Dialon and insulin
Pharmacology
- Sulfonylurea antidiabetic agent
- Stimulates insulin release from pancreatic beta cells
- Improves sensitivity of beta cells to glucose stimulus
- Reduces basal hepatic glucose production
- Increases peripheral tissue sensitivity to insulin
Dosage
- Initial dose of 1 mg once daily
- Dose titration based on regular blood sugar monitoring
- Usual dose range in well-controlled diabetes: 1 to 4 mg daily
- Timing and distribution of doses decided by physician
- Dose adjustment based on changes in weight or lifestyle
Administration
- Tablet must be swallowed with sufficient amount of liquid
Interaction
- Potentiates hypoglycemic action with certain drugs: ACE inhibitors, aminosalicylic acid, fibrates, fluconazole, etc.
- Attenuates hypoglycemic action with certain drugs: barbiturates, corticosteroids, diazoxide, etc.
- Beta-blockers, clonidine, and reserpine may lead to potentiation or weakening of the blood-glucose-lowering effect
- Acute and chronic alcohol intake may affect the activity of Dialon
Contraindications
- Not suitable for insulin-dependent diabetes mellitus
- Not suitable for diabetic ketoacidosis or diabetic coma
- Hypersensitivity to Glimepiride or other sulfonylureas
- Severe hepatic dysfunction and renal impairment
- Not for use in dialysis patients

Pregnancy & Lactation
- Not to be taken during pregnancy; changeover to insulin necessary

Precautions & Warnings
- Initial weeks of treatment may increase the risk of hypoglycemia
- Careful monitoring required
- Prompt control of hypoglycemia by immediate intake of carbohydrates
Overdose Effects
- Hypoglycemia
- Mild symptoms treated with oral glucose and dosage adjustments
- Severe reactions constitute medical emergencies
- Rapid intravenous injection of concentrated glucose solution for hypoglycemic coma
