Indications
- Adjunct to diet and exercise for lowering blood glucose in patients with noninsulin dependent diabetes mellitus (NIDDM)
- May be used with metformin or insulin
- Combination use with insulin may increase potential for hypoglycemia
Pharmacology
- Decreases blood glucose concentration
- Stimulates insulin release from pancreatic beta cells
- Improves sensitivity of beta cells to glucose stimulus
Dosage
- Initial dose usually 1 mg once daily
- Usual dose range for well-controlled diabetes is 1 to 4 mg daily
- Dose titration based on regular blood sugar monitoring
- Timing and distribution of doses decided by physician
Administration
- Must be swallowed with sufficient amount of liquid
Interaction
- Potentiates hypoglycemic action when used with certain drugs
- May be attenuated by other drugs
- Concomitant treatment with certain drugs may mask hypoglycemic symptoms
Contraindications
- Not suitable for insulin dependent diabetes mellitus or diabetic ketoacidosis
- Must not be used in patients hypersensitive to sulfonylureas or with severe hepatic/renal dysfunction

- Immediate intake of carbohydrates to promptly control hypoglycemia
Overdose Effects
- Can produce hypoglycemia
- Treatment includes oral glucose and adjustments in drug dosage or meal patterns
- Severe cases may require IV glucose injection and close monitoring
