Indications

  • Lowering blood glucose in noninsulin dependent diabetes mellitus (NIDDM)
  • Adjunct to diet and exercise for diabetes mellitus
  • Use in combination with insulin to lower blood glucose

Pharmacology

  • Primary mechanism of action: stimulating insulin release from pancreatic beta cells
  • Improving sensitivity of beta cells to glucose stimulus
  • Reducing basal hepatic glucose production
  • Increasing peripheral tissue sensitivity to insulin
  • Hypoglycaemic action persists for 24 hours

Dosage

  • Initial dose: 1 mg once daily
  • Usual dose range: 1 to 4 mg daily
  • Dose titration based on regular blood sugar monitoring
  • Timing and distribution of doses decided by physician
  • Swallow tablet with sufficient liquid

Administration

  • Tablet must be swallowed with sufficient liquid

Interaction

  • Drugs potentiating hypoglycaemic action of Limep
  • Drugs attenuating hypoglycaemic action of Limep
  • Potential for hypoglycaemia with concurrent use of insulin

Contraindications

  • Not suitable for insulin-dependent diabetes mellitus
  • Not for diabetic ketoacidosis or coma
  • Not for patients hypersensitive to Glimepiride or sulfonylureas
  • Not for patients with severe hepatic or renal dysfunction

Side Effects

  • Hypoglycaemia
  • Temporary visual impairment
  • Nausea, vomiting, diarrhoea
  • Abdominal pain
  • Urticaria
  • Fall in blood pressure

Pregnancy & Lactation

  • Not to be taken during pregnancy
  • Changeover to insulin necessary for pregnant patients
  • Not to be taken by breastfeeding women

Precautions & Warnings

  • Increased risk of hypoglycaemia in initial weeks of treatment
  • Necessity for dosage adjustment in the presence of hypoglycaemia

Overdose Effects

  • Hypoglycaemic symptoms without loss of consciousness
  • Severe hypoglycaemic reactions with coma or seizure
  • Immediate treatment with oral glucose or rapid intravenous injection of concentrated glucose solution
