Indications
- Lowering blood glucose in noninsulin dependent diabetes mellitus (NIDDM)
- Adjunct to diet and exercise for diabetes mellitus
- Use in combination with insulin to lower blood glucose
Pharmacology
- Primary mechanism of action: stimulating insulin release from pancreatic beta cells
- Improving sensitivity of beta cells to glucose stimulus
- Reducing basal hepatic glucose production
- Increasing peripheral tissue sensitivity to insulin
- Hypoglycaemic action persists for 24 hours
Dosage
- Initial dose: 1 mg once daily
- Usual dose range: 1 to 4 mg daily
- Dose titration based on regular blood sugar monitoring
- Timing and distribution of doses decided by physician
- Swallow tablet with sufficient liquid
Administration
- Tablet must be swallowed with sufficient liquid
Interaction
- Drugs potentiating hypoglycaemic action of Limep
- Drugs attenuating hypoglycaemic action of Limep
- Potential for hypoglycaemia with concurrent use of insulin
Contraindications
- Not suitable for insulin-dependent diabetes mellitus
- Not for diabetic ketoacidosis or coma
- Not for patients hypersensitive to Glimepiride or sulfonylureas
- Not for patients with severe hepatic or renal dysfunction
Side Effects
- Hypoglycaemia
- Temporary visual impairment
- Nausea, vomiting, diarrhoea
- Abdominal pain
- Urticaria
- Fall in blood pressure
Pregnancy & Lactation
- Not to be taken during pregnancy
- Changeover to insulin necessary for pregnant patients
- Not to be taken by breastfeeding women
Precautions & Warnings
- Increased risk of hypoglycaemia in initial weeks of treatment
- Necessity for dosage adjustment in the presence of hypoglycaemia
Overdose Effects
- Hypoglycaemic symptoms without loss of consciousness
- Severe hypoglycaemic reactions with coma or seizure
- Immediate treatment with oral glucose or rapid intravenous injection of concentrated glucose solution
