Description

  • Indications:
    • Adjunct to diet and exercise for lowering blood glucose in noninsulin dependent diabetes mellitus (NIDDM)
    • Can be used in combination with metformin or insulin for glycaemic control
  • Pharmacology:
    • Stimulates insulin release from pancreatic beta cells
    • Reduces hepatic glucose production
    • Increases peripheral tissue sensitivity to insulin
  • Dosage:
    • Initial dose is 1 mg once daily, can be titrated up to 6 mg based on glucose monitoring
    • Timing and distribution of doses decided by physician based on patient's lifestyle
    • May require dose adjustment with changes in weight or lifestyle
  • Administration:
    • Tablet to be swallowed with sufficient liquid
  • Interaction:
    • Potentiating drugs include ACE inhibitors, aminosalicylic acid, and others
    • Attenuating drugs include barbiturates, corticosteroids, and others
  • Contraindications:
    • Not suitable for insulin dependent diabetes mellitus or diabetic coma
    • Not for use in patients hypersensitive to Glimepiride or with severe hepatic or renal dysfunction

    • Hypoglycaemia

  • Pregnancy & Lactation:
    • Not to be taken during pregnancy, changeover to insulin is necessary

    • Initial weeks of treatment may increase risk of hypoglycaemia
    • Prompt control of hypoglycaemia with immediate intake of carbohydrates
  • Overdose Effects:
    • Overdosage can produce hypoglycaemia
    • Mild hypoglycaemic symptoms should be treated aggressively with oral glucose
    • Severe reactions require hospitalization and rapid intravenous glucose
