Indications
- Adjunct to diet and exercise for lowering blood glucose in Type II diabetes mellitus
- May be used with metformin or insulin for glycaemic control
Pharmacology
- Sulfonylurea antidiabetic agent
- Stimulates insulin release from pancreatic beta cells
- Reduces basal hepatic glucose production
- Increases peripheral tissue sensitivity to insulin
Dosage
- Initial dose of 1mg once daily
- Usual dose range in well-controlled diabetes is 1 to 4mg daily
- Timing and distribution of doses decided by physician
- Secondary dosage adjustment based on improved diabetes control and other factors
Administration
Tablet must be swallowed with sufficient liquid

Contraindications
- Insulin-dependent diabetes mellitus
- Diabetic ketoacidosis
- Hypersensitivity to Glimepiride or sulfonylureas
- Severe hepatic or renal dysfunction

- Temporary visual impairment
- Nausea
- Vomiting
- Diarrhoea
- Abdominal pain
- Urticaria
- Fall in blood pressure
Pregnancy & Lactation
- Contraindicated during pregnancy
- Changeover to insulin needed
- Harmful during breastfeeding
Precautions & Warnings
- Increased risk of hypoglycaemia in initial weeks of treatment
- Immediate intake of carbohydrates for hypoglycaemia control
Overdose Effects
- Hypoglycaemic symptoms without loss of consciousness
- Severe hypoglycaemic reactions with coma or seizure
- Need for aggressive treatment with oral glucose or IV injection of glucose
