Indications
- Adjunct to diet for controlling hyperglycaemia
- Treatment of non-insulin-dependent diabetes mellitus
- Reduction in daily insulin dose
Pharmacology
- Stimulates insulin release from pancreas
- Increases insulin sensitivity
- Decreases hepatic glucose production
Dosage & Administration
- Initial dose of 5mg before breakfast
- Dosage adjustments based on blood glucose response
- Maintenance dose ranging from 2.5 to 30mg daily
- Administration approximately 30 minutes before a meal
- Dosing considerations for elderly, debilitated, or malnourished patients
Interaction
- Hypoglycaemic action potentiated by certain drugs
- Drugs that may lead to loss of control on blood glucose
Contraindications
- Hypersensitivity to Glipizide or its components
- Juvenile onset diabetes
- Severe or unstable 'brittle' diabetes
- Diabetes complicated by ketosis and acidosis
- Severe renal, hepatic, or thyroid impairment
Side Effects
- Dose related gastrointestinal complaints
- Allergic skin reactions
- Haematologic adverse effects
- Metabolic and endocrine reactions
- Dizziness, drowsiness, and headache
Pregnancy & Lactation
- Use during pregnancy if potential benefit justifies potential risk
- Prolonged severe hypoglycaemia in neonates
- Discontinuation one month before expected delivery date
- Not recommended during lactation
Precautions & Warnings
- Capability of producing severe hypoglycaemia
- Patient selection, dosage, and instructions to avoid hypoglycaemic episodes
- Susceptibility in elderly, debilitated, or malnourished patients
- Slow metabolism and excretion in patients with impaired renal and/or hepatic function
- Necessity of discontinuing and administering insulin during stress
- Considerations for patients experiencing loss of control on blood glucose
