Indications
- Adjunct to diet for controlling hyperglycaemia
- Treatment of non-insulin-dependent diabetes mellitus
- Reduction in daily dose of insulin
Pharmacology
- Stimulates insulin release from the pancreas
- Increases insulin sensitivity
- Decreases hepatic glucose production
Dosage & Administration
- Initial dose of 5 mg before breakfast
- Dosage adjustments by increment of 2.5-5 mg
- Maximum recommended daily dose of 40 mg
Interaction
- Potentiation of hypoglycaemic action by certain drugs
- Potential hyperglycaemic effects of certain drugs
- Observation for hypoglycaemia or hyperglycaemia when co-administered or withdrawn
Contraindications
- Hypersensitivity to Glipizide
- Juvenile onset diabetes
- Severe or unstable ‘brittle’ diabetes
- Severe renal, hepatic, or thyroid impairment
Side Effects
- Dose-related gastrointestinal complaints
- Allergic skin reactions
- Reported haematologic and metabolic side effects
Pregnancy & Lactation
- Use during pregnancy only if potential benefit justifies potential risk
- Recommended discontinuation at least one month before expected delivery date
- Not recommended during lactation
Precautions & Warnings
- Capability of producing severe hypoglycaemia
- Proper patient selection, dosage, and instructions to avoid hypoglycaemic episodes
- Particular susceptibility in elderly, debilitated, or malnourished patients
