Therapeutic Class

Indications

  • Control of hyperglycaemia
  • Non-insulin-dependent diabetes mellitus treatment
  • Adjunct to diet

Pharmacology

  • Stimulates insulin release
  • Increases insulin sensitivity
  • Decreases hepatic glucose production

Dosage & Administration

  • Initial dose: 5 mg before breakfast
  • Dosage adjustments based on blood glucose response
  • Maximum recommended once daily dose: 15 mg
  • Maximum recommended total daily dose: 40 mg
  • Administered approximately 30 minutes before a meal
  • Special dosage considerations for geriatric patients, those with liver disease, and impaired renal or hepatic function

Interaction

  • Potentiation of hypoglycaemic action by certain drugs
  • Hyperglycaemic effects of certain drugs
  • Caution when co-administered with specific drug classes

Contraindications

  • Hypersensitivity to Glipizide or components
  • Juvenile onset diabetes
  • Severe or unstable 'brittle' diabetes
  • Diabetes complicated by ketosis and acidosis, major surgery, severe sepsis or severe trauma
  • Severe renal, hepatic or thyroid impairment, co-existent renal and hepatic disease

Pregnancy & Lactation

  • Potential risk to the fetus

  • Not recommended during breastfeeding

  • Severe hypoglycaemia risk
  • Proper patient selection and dosage
  • Increased risk in elderly, debilitated, or malnourished patients
  • Metabolism and excretion considerations in patients with impaired renal and/or hepatic function
  • Possibility of loss of control on blood glucose under stress
  • Diminished drug responsiveness
