Indications

Pharmacology

  • Biguanide type oral antihyperglycemic drug
  • Lowers both basal and postprandial plasma glucose
  • Different mechanism of action from sulfonylureas
  • Does not produce hypoglycemia
  • Decreases hepatic glucose production
  • Improves insulin sensitivity
  • Decreases intestinal absorption of glucose

Contraindications

  • Hypersensitivity to active substance
  • Acute metabolic acidosis
  • Severe renal failure
  • Acute conditions altering renal function
  • Conditions causing tissue hypoxia

Side Effects

  • Hemolytic anemia
  • Lactic acidosis
  • Decrease of vitamin B12 absorption
  • Peripheral neuropathy with vitamin B12 deficiency
  • Taste disturbance
  • Gastrointestinal disorders
  • Liver function test abnormalities
  • Skin reactions

Pregnancy & Lactation

  • Increased risk of congenital abnormalities
  • Use of insulin recommended during pregnancy
  • Excretion into human breast milk

Precautions & Warnings

  • Excretion by the kidney
  • Lowering of vitamin B12 level
  • Increase risk of hypoglycemia in combination with insulin
