Indications
- Treatment of type 2 diabetes mellitus in adults
- Treatment of type 2 diabetes mellitus in children from 10 years of age and adolescents
- Reduction of diabetic complications in overweight type 2 diabetic adult patients
Pharmacology
- Lowering of basal and postprandial plasma glucose
- Decrease in hepatic glucose production
- Improvement in insulin sensitivity
Dosage & Administration
- Usual starting dose of 500 mg for adults
- Usual starting dose of 500 mg for children
- Dosage increases should be made in increments of 500 mg weekly
Interaction
- Enhancement of Ormin absorption by Nifedipine
- Potential interaction with cationic drugs
- Certain drugs may lead to loss of glycemic control
Contraindications
- Hypersensitivity to the active substance or excipients
- Any type of acute metabolic acidosis
- Severe renal failure (GFR <30 mL/min)

Pregnancy & Lactation
- Increased risk of congenital abnormalities and perinatal mortality with uncontrolled diabetes during pregnancy

Precautions & Warnings
- Substantial excretion by the kidney
- Lowering of vitamin B12 level
- Increased risk of hypoglycemia in combination with insulin
- Need for regular monitoring of renal function
Use in Special Populations
- Dosage adjustment based on renal function in elderly subjects
- Careful follow-up of the effect on growth and puberty in metformin-treated children
Overdose Effects
- Lactic acidosis as a medical emergency
- Hemodialysis as the most effective method of treatment
