Indications

Description

  • Oral blood-glucose-lowering drug
  • Belongs to the meglitinide class
  • Works by causing pancreas to release more insulin into the bloodstream
  • Rapid onset of action and elimination

Pharmacology

  • Binds to specific receptors in the cell membrane
  • Leads to closure of ATP dependent K+ channels
  • Depolarization of cell membrane
  • Ca++ influx
  • Increased intracellular Ca++
  • Stimulation of insulin secretion

Dosage & Administration

  • Starting dose based on HbA1c levels
  • Should be taken before each meal
  • Dosage adjustment according to response
  • Maximum dosage of 16 mg daily

Contraindications

Overdose Effects

  • Few adverse effects with doses up to 80 mg
  • Hypoglycemic reactions with coma, seizure, or neurological impairment
