Pharmacology
- Binds to specific receptors in cell membrane
- Closure of ATP dependent K+ channels
- Depolarisation of cell membrane
- Ca++ influx
- Stimulation of insulin secretion
Dosage & Administration
- Starting dose 0.5 mg before each meal
- Dosage adjustment according to response
- Maximum 16 mg daily
- Taken immediately or up to 30 minutes before each meal
Interaction
- Cytochrome P450 enzyme system
- Inhibitors and inducers of metabolism
- Effects on other drugs such as Digoxin, Theophyllin, and Warfarin
- Increased risk of hypoglycaemia with certain drugs
Contraindications
- Diabetic ketoacidosis
- Type 1 diabetes mellitus
- Known hypersensitivity to the drug or its inactive ingredients

Precautions & Warnings
- Substitution of insulin during concurrent illness and surgery
- Capable of producing hypoglycemia
- Administration with meals to lessen the risk of hypoglycemia
Overdose Effects
- Few adverse effects other than lowering of blood glucose up to 80 mg
- Severe hypoglycemic reactions occur infrequently
