Indications
- Adjunct to diet and exercise for type II diabetes
- Monotherapy and combination therapy indication
- Improves glycaemic control
- Use in combination with sulphonylurea, Metformin or Insulin
Pharmacology
- Belongs to thiazolidinediones class
- Depends on the presence of Insulin for mechanism of action
- Decreases Insulin resistance in the periphery and liver
- Activates peroxisome proliferator activated receptor gamma (PPAR-γ)
Dosage & Administration
- Can be taken once daily without regard to meals
- Individualized management
- Initiate at 15 mg or 30 mg once daily for monotherapy
- Maximum recommended daily dose is 45 mg
- Consider combination therapy for inadequate response
Interaction
- Reduces the plasma concentration of oral contraceptives containing ethinyl oestradiol and norethindrone
Contraindications
- Contraindicated in patients with known hypersensitivity to any components
Side Effects
- Incidence of adverse events in clinical trials

- Incidence of upper respiratory tract infection, headache, sinusitis, myalgia, tooth disorder, and pharyngitis
Pregnancy & Lactation
- Use during pregnancy if potential benefit justifies potential risk to the foetus
- Unknown secretion in human milk
- Should not be administered to lactating women
Precautions & Warnings
- Exerts antihyperglycaemic effect only in the presence of Insulin
- Should not be used in Type 1 diabetes or diabetic ketoacidosis
- Use with caution in case of combination antidiabetic therapy and hepatic insufficiency
- Regular monitoring of liver enzymes
