Indications
- Replacement therapy in hypothyroidism
- Suppression of TSH levels

Pharmacology
- Contains synthetic Levothyroxine
- Identical to natural hormone T4
- Converted to T3 in peripheral tissues
- Long half-life in circulation
- Major carrier of T4 is TBG
- 30% converted to T3
- Enterohepatic circulation
Dosage & Administration
- Adult dose starting from 25-50 mcg/day
- Increase in dose at 6-8 week intervals
- Pediatric dosage based on age and clinical response
- Dose adjustments based on clinical response and lab parameters
Interaction
- Concurrent use with tri/tetracyclic antidepressants may increase therapeutic and toxic effects
- Addition to antidiabetic or insulin therapy may increase requirements
- Reduction in serum digitalis glycoside levels in hyperthyroidism
Contraindications
- Untreated Thyrotoxicosis
- Acute Myocardial Infarction
- Uncorrected Adrenal failure
Side Effects

Pregnancy & Lactation

Precautions & Warnings
- Adrenocortical insufficiency in pituitary gland function hypothyroidism
- Care required in patients with cardiovascular disease and severe hypothyroidism
- Adjustment of dosage with marked change in body weight
- Monitoring blood levels of T3 and T4 for correct dosage
