Pharmacology

Synthetic adrenocortical drug with predominantly glucocorticoid properties, inhibits Phospholipase A2 enzyme, rapidly absorbed from the GI tract, metabolized in the liver, excreted in the urine

Dosage & Administration

  • Adult:
    • Nephrotic Syndrome:
      • Initial: 2 mg/kg/day
      • Maintenance dose: 0.5 to 1 mg/kg
    • Anti-inflammatory: 5 to 60 mg per day
    • Acute Asthma: 40-60 mg/day
    • Allergic Conditions:
      • Day 1: 10 mg PO
      • Day 2: 5 mg PO
      • Day 3: 5 mg PO
      • Day 4: 5 mg PO
      • Day 5: 5 mg PO
      • Day 6: 5 mg PO
  • Pediatric:
    • Asthma:
      • 1 year: Acute: 10 mg
      • Maintenance: 10 mg
      • 1 to 4 years: Acute: 20 mg
      • Maintenance: 20 mg
      • 5 to 12 years: Acute: 30 mg
      • Maintenance: 30 mg
      • 12 years: Acute: 40 mg
      • Maintenance: 40 mg
    • Anti-inflammatory: 0.05 to 2 mg/kg/day
    • Immunosuppression: 0.05 to 2 mg/kg/day

Interaction

Efficacy reduced by Aminoglutethimide, Antacids, Barbiturates, Carbamazepine, Griseofulvin, Mitotane, Phenylbutazone, Phenytoin, Primidone, Rifampin. Reduces potassium levels. Immunization precaution.

Contraindications

Systemic infections, hypersensitivity to any ingredient, ocular herpes simplex

Overdose Effects

Adverse effects develop after prolonged use, treatment is symptomatic, reduce dose gradually
