Description
A synthetic adrenocortical drug used in the treatment of various conditions such as rheumatic disorders, endocrine disorders, dermatologic diseases, allergic states, respiratory diseases, hematologic disorders, edematous states, gastrointestinal diseases, nephrotic syndrome, asthma, and immunosuppression.

Pharmacology
Prednisolone is a synthetic adrenocortical drug with predominantly glucocorticoid properties. It inhibits the action of the Phospholipase A2 enzyme responsible for producing inflammatory mediators. Well absorbed from the gastrointestinal tract with a half-life of 2 to 4 hours. Metabolized in the liver and excreted in the urine.
DosageAndAdministration
- Adult:
- NephroticSyndrome: 2 mg/kg/day in divided doses; maintenance: 0.5 to 1 mg/kg every other day
- Anti-inflammatory: 5 to 60 mg per day in divided doses
- AcuteAsthma: 40-60 mg/day PO in single daily dose or divided q12 hr for 3-10 days
- AllergicConditions: Gradual tapering from 10 mg to 5 mg over 6 days
- Pediatric:
- Asthma: 10-40 mg orally every 12 hours depending on age
- Anti-inflammatory: 0.05 to 2 mg/kg/day divided 1 to 4 times/day
- Immunosuppression: 0.05 to 2 mg/kg/day divided 1 to 4 times/day

Systemic infections, hypersensitivity to any ingredient, ocular herpes simplex

PrecautionsAndWarnings
Caution in diabetes, hypertension, psychological disturbances, osteoporosis, pregnancy, chronic nephritis. Long-term use can cause Cushing's habitus, hyperglycemia, muscular weakness, increased susceptibility to infection, delayed wound healing, psychiatric disturbances.
OverdoseEffects
Adverse effects develop after prolonged use of excessive doses. Treatment is symptomatic with gradual dose reduction.
