Description

- Used in selected cases of endometrial carcinoma and benign breast disease

- Postponement of menstruation
- As an adjunct to estrogen therapy in pre-and postmenopause in order to avoid endometrial hyperplasia
- Lynestrenol is the synthetic progestagen in Orgatril
- Lynestrenol has progestational influence on the endometrium
- Suppresses ovulation and menstruation when taken daily without interruption
- Lynestrenol is readily absorbed and converted into the pharmacologically active norethisterone
- Peak plasma levels of norethisterone are reached within 2-4 hours after ingestion
- Lynestrenol and its metabolites are predominantly excreted in the urine
- Lynestrenol tablets should be taken orally with some fluid
- Interactions with activated charcoal, barbiturates, hydantoins, rifampicin, beta-adrenergic blockers, cyclosporin, and insulin
- Contraindicated during pregnancy or suspected pregnancy
- Contraindicated in severe liver disease, undiagnosed vaginal bleeding, and conditions affected by sex steroids
- Frequent side effects include breakthrough bleeding, change in libido, nausea, gastrointestinal disturbances, and weight increase
- Occasional side effects include headache, migraine, dizziness, nervousness, depression, increased sweating, acne, hirsutism, chloasma, rash, pruritus, jaundice, alterations in liver function tests, menstrual irregularity, and reduced glucose tolerance
- Chloasma occasionally seen during use, especially in women with a history of chloasma gravidarum
- Changes in parameters of liver function, carbohydrate metabolism, and haemostasis may occur
- During prolonged treatment with progestagens, periodic medical examinations are advisable
- Weak androgenic activity confirmed by slight signs of virilisation observed in some patients
- Close medical supervision for patients with circulatory disorders, severe depression, and other specific conditions
- Stored in a cool & dry place, away from light, and out of the reach of children
Dosage schedule
- Polymenorrhoea: 1 tablet daily on days 14-25 of the cycle
- Premenstrual syndrome: 1 tablet daily on days 4-25 of the cycle
- Endometriosis: 1-2 tablets daily for at least 6 months
- Selected cases of endometrial carcinoma: 6-10 tablets daily for prolonged periods
- Benign breast disease: 1 tablet daily on days 14-25 of the cycle for at least 3-4 months
- Menorrhagia and metrorrhagia: 1 tablet daily on days 14-25 of the cycle, repeated for the next 3 menstrual cycles
- Selected cases of primary and secondary amenorrhoea and oligomenorrhoea: Treatment with estrogen in conjunction with 1 tablet daily of Lynestrenol on days 14-25
- Suppression of menstruation ovulation and ovulation pain dysmenorrhoea: Treatment with 1 tablet daily, can be continued for many months without tablet-free days
- Postponement of menstruation: Treatment with 1 tablet daily starting 2 weeks before the expected onset of menstruation
- As an adjunct to estrogen therapy in pre-and postmenopause in order to avoid endometrial hyperplasia: 1/2-1 tablet daily for 12-15 days per month
