Indications

  • Treatment of secondary amenorrhea
  • Treatment of abnormal uterine bleeding
  • Reduction of endometrial hyperplasia
  • Indicated for nonhysterectomized postmenopausal women with daily oral conjugated estrogens

Pharmacology

  • Transforms proliferative into secretory endometrium
  • Androgenic and anabolic effects
  • Inhibits gonadotropin production
  • Devoid of significant estrogenic activity

Dosage & Administration

  • Dosage of 5 or 10 mg daily for 5-10 days for secondary amenorrhea
  • Dosage of 5 or 10 mg daily for 5-10 days for abnormal uterine bleeding
  • Dosage of 5 or 10 mg daily for reduction of endometrial hyperplasia in postmenopausal women
  • Consideration for planned menstrual cycling

Contraindications

  • Undiagnosed abnormal genital bleeding
  • History of breast cancer
  • Estrogen- or progesterone-dependent neoplasia
  • Active DVT, PE, or arterial thromboembolic disease
  • Known anaphylactic reaction to Medroxyprogesterone Acetate
  • Liver impairment or disease
  • Pregnancy

  • Breast tenderness
  • Thromboembolic disorders
  • Nausea
  • Skin sensitivity reactions
  • Neuro-ocular lesions
  • Mental depression

Pregnancy & Lactation

  • Should not be used during pregnancy
  • Should not be used during lactation
  • Detectable amounts identified in breast milk

Precautions & Warnings

  • Addition of a progestin when a woman has not had a hysterectomy
  • Monitoring of unexpected abnormal vaginal bleeding
  • Regular monitoring of blood pressure
  • Consideration for hypertriglyceridemia
  • Special caution in cases of hepatic impairment
  • Observation for fluid retention
  • Caution in women with hypoparathyroidism
  • Consideration for exacerbation of other conditions

Use in Special Populations

  • Not indicated in children (pediatric use)
  • Insufficient data for geriatric use

Overdose Effects

  • Nausea and vomiting
  • Breast tenderness
  • Dizziness
  • Abdominal pain
  • Withdrawal bleeding
