Indications
- Treatment of anovulation in females
- Treatment of oligo-ovulation in females
- Treatment of infertility in males with oligospermia
- Treatment of infertility in males with poor sperm motility
- Treatment of lactational amenorrhoea
- Treatment of psychogenic amenorrhoea
- Treatment of post-oral contraceptive amenorrhoea
- Treatment of secondary amenorrhoea of unknown origin
Pharmacology
- Mainly antiestrogenic effects
- Estrogenic effects
- Stimulates ovulation
- Increases FSH and LH secretion
- Increases ovarian follicle maturation
- Triggers preovulatory LH surge
- Promotes corpus luteum development
- Likely related to increases in FSH and LH secretion in male infertility
Dosage & Administration
- 50 mg daily for 5 days for females
- Starts any day from second to fifth day of the menstrual cycle if bleeding occurs
- Smaller dose for individual unusually sensitive to clomiphene
- 75 mg to 100 mg daily for five days if ovulation does not occur
- Three courses of therapy to assess ovulation
- 25 mg once daily up to 90 days for males
- Break of 5 days after every 25 days for males
- Loading dose of 250 mg-1 g for patients undergoing haemodialysis
- Elderly dosage same as adults
- Ovulation usually occurs 4 to 10 days after the last day of treatment

- Pregnancy
- Liver insufficiency
- Abnormal vaginal bleeding (uterine tumour, ovarian cyst)
