Indications
- Endometriosis

Pharmacology
- Gonadotropin inhibitor
- Pituitary-ovarian axis suppression
- Inhibition of pituitary gonadotropins output
- Suppression of ovarian estrogen production
- Suppression of ovarian function
- Androgen, progesterone, and glucocorticoid receptor binding
- Effect on steroid receptor sites in breast tissue
- Increase in the metabolic clearance rate of progesterone
Dosage & Administration
- Given in up to 4 divided doses
- Treatment should start during menstruation
- Daily doses ranging from 100mg to 800mg
- Adjusted to achieve amenorrhea
- Duration of treatment from 3-9 months
- Not recommended for use in children

- Potential interaction with insulin
- Potential interaction with estrogens and progestogens
- Potential interaction with anticonvulsant therapy
- Potential interaction with antihypertensive therapy

Contraindications
- Pregnancy and breast-feeding
- Impaired hepatic, renal, or cardiac function
- Porphyria
- Thromboembolic disease
- Androgen-dependent tumor
- Abnormal vaginal bleeding that has not been fully investigated
- Hypersensitivity to danazol

- Visual disturbances
