Pharmacology

  • Gonadotropin inhibitor
  • Pituitary-ovarian axis suppression
  • Inhibition of pituitary gonadotropins output
  • Suppression of ovarian estrogen production
  • Suppression of ovarian function
  • Androgen, progesterone, and glucocorticoid receptor binding
  • Effect on steroid receptor sites in breast tissue
  • Increase in the metabolic clearance rate of progesterone

Dosage & Administration

  • Given in up to 4 divided doses
  • Treatment should start during menstruation
  • Daily doses ranging from 100mg to 800mg
  • Adjusted to achieve amenorrhea
  • Duration of treatment from 3-9 months
  • Not recommended for use in children

  • Potential interaction with insulin
  • Potential interaction with estrogens and progestogens
  • Potential interaction with anticonvulsant therapy

Contraindications

  • Pregnancy and breast-feeding
  • Impaired hepatic, renal, or cardiac function
  • Porphyria
  • Thromboembolic disease
  • Androgen-dependent tumor
  • Abnormal vaginal bleeding that has not been fully investigated
  • Hypersensitivity to danazol
