Description
- Suppression of pituitary-ovarian axis
- Inhibition of FSH and LH output
- Suppression of ovarian estrogen production
- Inhibition of ovarian steroidogenesis
- Binding to androgen, progesterone, and glucocorticoid receptors
- Increase in metabolic clearance rate of progesterone
- Suppression of ovarian function leading to anovulation and amenorrhea
- Correction of underlying biochemical deficiency for hereditary angioedema
Dosage
- Dosing up to 4 divided doses
- Start during menstruation in women of child-bearing potential
- 200-800 mg daily for endometriosis
- 100-200 mg daily for menorrhagia
- 100-400 mg daily for severe cyclical mastalgia
- 300 mg daily for benign breast cysts
- 400 mg daily for gynaecomastia
- 400-800 mg daily for pre-operative thinning of endometrium
- Not used in children
Interaction
- Increased insulin resistance
- Interaction with exogenous estrogens and/or progestogens
- Effect on plasma concentration of anticonvulsant and antihypertensive therapy
- Increase in plasma concentration of cyclosporine
- Provocation of migraine and potential reduction in effectiveness of migraine therapy
Contraindications
- Pregnancy and breast-feeding
- Impaired hepatic, renal, or cardiac function
- Porphyria
- Thromboembolic disease
- Androgen-dependent tumor
- Abnormal vaginal bleeding
- Hypersensitivity to danazol

- Musculo-skeletal spasm

- Temporary alteration in lipoproteins
- Headache and benign intracranial hypertension
Pregnancy and Lactation
- Risk of virilization of female fetus
- Avoid in pregnant women and during breastfeeding
Precautions and Warnings
- Cardiac, hepatic or renal impairment
- Elderly
- Polycythaemia
- Epilepsy
- Diabetes mellitus
- Hypertension
- History of thrombosis or thromboembolic disease
- Immediate cessation if signs of virilization occur
