Indications
- Endometriosis
- Benign breast disease
- Menorrhagia
- Gynecomastia
- Preoperative thinning of the endometrium before hysteroscopic endometrial ablation.
Pharmacology
- Gonadotropin inhibition
- Suppression of pituitary-ovarian axis
- Suppression of ovarian estrogen production
- Inhibition of ovarian steroidogenesis
- Binding to androgen, progesterone, and glucocorticoid receptors
- Increase in the metabolic clearance rate of progesterone
Dosage & Administration
- Given in up to 4 divided doses
- Treatment should start during menstruation
- Daily doses adjusted to achieve amenorrhoea
- Specific dosages for different indications
Interaction
- Increased insulin resistance
- Potential interaction with exogenous estrogens and/or progestogens
- Possible effects on anticonvulsant therapy
- Potential opposition to antihypertensive agents
- Potential increase in the plasma concentration of cyclosporine
Contraindications
- Pregnancy and breast-feeding
- Impaired hepatic, renal, or cardiac function
- Porphyria
- Thromboembolic disease
- Androgen-dependent tumor
- Abnormal vaginal bleeding
- Hypersensitivity to danazol

Pregnancy & Lactation
- Risk of virilization of a female fetus
- Drug should not be given to pregnant women
- Patients taking the drug should not breast-feed

- Cardiac, hepatic or renal impairment
- Polycythaemia
- Epilepsy
- Diabetes mellitus
- Hypertension
- Migraine
- History of thrombosis or thromboembolic disease
- Signs of virilization
