Indications

Description
- Selective, reversible inhibitor of cyclic guanosine monophosphate (cGMP)-specific phosphodiesterase type 5 (PDE5)
- Causes smooth muscle relaxation and inflow of blood into the penile tissues
Mode of Action
- Enhances the effect of nitric oxide (NO) by inhibiting phosphodiesterase type 5 (PDE5)
- Rapidly absorbed after oral administration
- Metabolized predominantly by hepatic cytochrome P450 3A4
Dosage & Administration
- Recommended dose: 50 mg taken 1 hour before sexual activity
- Can be taken anywhere from 4 hours to 0.5 hour before sexual activity
Interaction
- Metabolism mediated by cytochrome P450 isoforms 3A4 and 2C9
- Inhibitors or inducers of these isoenzymes may affect clearance of the medication
Contraindications
- Hypersensitivity to any component of the medication
- Concomitant use of nitrates
Side Effects
- Body as a whole: face edema, chills, abdominal pain
- Cardiovascular: hypotension, myocardial ischemia, heart failure
- Digestive: vomiting, dry mouth
- Nervous: insomnia, depression
- Respiratory: dyspnea, cough increased
Pregnancy & Lactation
- Pregnancy category B
- Not indicated for use by women
Precautions & Warnings
- Caution advised when co-administered with alpha-blockers
- Consideration for patients with underlying cardiovascular disease
Overdose Effects
- Adverse events similar to lower doses but with increased incidence rates and severities
