Indications

Description
- Selective, reversible inhibitor of cyclic guanosine monophosphate (cGMP)-specific phosphodiesterase type 5 (PDE5)
- Causes smooth muscle relaxation and increase in blood flow to penile tissues
Pharmacology
- Mechanism of Action involves release of nitric oxide (NO) in the corpus cavernosum during sexual stimulation
- Rapid absorption after oral administration with mean absolute bioavailability of 41%
- Metabolized predominantly by hepatic metabolism
- Maximum observed plasma concentrations reached within 30 to 120 minutes of oral dosing
Dosage & Administration
- Recommended initial dose of 50 mg, approximately 1 hour before sexual activity
- Dosing frequency is once per day
- Dose may be adjusted based on effectiveness and toleration
Interaction
- Metabolism is mediated by cytochrome P450 (CYP) isoforms 3A4 and 2C9
- Inhibitors of these isoenzymes may reduce clearance, while inducers may increase clearance
Contraindications
- Hypersensitivity to any component of the medication
- Contraindicated in patients using organic nitrates
