Description

Eurofil is a selective, reversible inhibitor of cyclic guanosine monophosphate (cGMP)-specific phosphodiesterase type 5 (PDE5).

Mechanism of Action

Inhibition of PDE5 produces increased levels of cGMP in the corpus cavernosum, resulting in smooth muscle relaxation and inflow of blood into the penile tissues.

Pharmacokinetics and Metabolism

Mean absolute bioavailability of 41%
Eliminated predominantly by hepatic metabolism
Terminal half lives of about 4 hours

Dosage & Administration

Recommended dose of 50 mg
Dose may be increased to a maximum recommended dose of 100 mg or decreased to 25 mg
Maximum recommended dosing frequency is once per day

Interaction

Metabolism mediated by CYP isoforms 3A4 and 2C9
Inhibitors of these isoenzymes may reduce Eurofil clearance

Contraindications

Hypersensitivity to any component
Use of organic nitrates

Precautions & Warnings

Co-administration with alpha-blockers
