Indications
- Treatment of erectile dysfunction in men
- Treatment of pulmonary arterial hypertension
Dosage & Administration
- Recommended starting dose of 50mg
- Can be taken 1 hour before sexual activity
- Maximum recommended dose of 100mg
- Maximum dosing frequency is once per day
- Recommended dose for pulmonary arterial hypertension is 20mg three times a day
Interaction
- Metabolism mediated by cytochrome P450 isoforms 3A4 and 2C9
- Inhibitors of these isoenzymes may reduce clearance
- Inducers of these isoenzymes may increase clearance
- Different drugs can impact Vigorex systemic exposure
Contraindications
- Hypersensitivity to any component of the medication
- Use of organic nitrates

Precautions & Warnings
- Evaluation of erectile dysfunction should include determination of potential underlying causes
- Caution advised when co-administered with alpha-blockers
- Potential for additive effect on blood pressure when co-administered with vasodilators
- Considerations for patients on alpha-blocker therapy and PDE5 inhibitors
- Potential cardiac risk in patients with preexisting cardiovascular disease
- Vigorex has vasodilatory properties
- Patients sensitive to actions of vasodilators should be cautious
- No controlled clinical data on safety or efficacy in certain patient groups
- Potential for prolonged erection and priapism
- Caution with use in patients taking ritonavir
- Visual disturbances reported at higher levels of exposure
- Decrease in dosage recommended for patients taking ritonavir
Overdose Effects
- Adverse events similar to those seen at lower doses
- Incidence rates and severities increased at higher doses
- Standard supportive measures should be adopted in cases of overdose
- Renal dialysis not expected to accelerate clearance
