Description

Selective, reversible inhibitor of phosphodiesterase type 5 (PDE5)
Causes smooth muscle relaxation and inflow of blood into penile tissues during sexual stimulation

Mechanism of Action

Involves release of nitric oxide during sexual stimulation
Activates enzyme guanylate cyclase
Increases levels of cyclic guanosine monophosphate (cGMP)

Bioavailability

Mean absolute bioavailability of 41%

Metabolism

Eliminated predominantly by hepatic metabolism
Converted to an active metabolite with properties similar to the parent

Maximum Plasma Concentrations

Reached within 30 to 120 minutes of oral dosing

Pharmacokinetics in Special Populations

Healthy elderly volunteers had a reduced clearance of Sildenafil

Recommended Dosage for Erectile Dysfunction

50 mg taken approximately 1 hour before sexual activity
May be increased to a maximum recommended dose of 100 mg or decreased to 25 mg based on toleration
