Administration Guidelines

Take with meals for maximum absorption, or one hour before or two hours after meals for best results

Dosage for Adults

250-500 mg every 6 hours

Dosage for Children

  • 125-250 mg every 6 hours
  • 125 mg/5 ml powder for suspension: 1-2 teaspoonful (5-10 ml) every 6 hours
  • 250 mg/5 ml powder for suspension: ½-1 teaspoonful (2.5-5 ml) every 6 hours

Dosage for Infants

  • 62.5-125 mg every 6 hours
  • 125 mg/5 ml powder for suspension: ½-1 teaspoonful (2.5-5 ml) every 6 hours, or as prescribed by the physician

Best Time for Administration

Best taken on an empty stomach, preferably one hour before meals

Food Interaction

Concurrent intake of food reduces absorption rate

Drug Interactions

  • Increased levels of methotrexate in serum
  • Disturbed absorption of other orally administered drugs if diarrhea occurs
  • Attenuated activity if combined with bacteriostatic chemotherapeutics or antibiotics
  • Renal excretion inhibition when combined with probenecid
  • Elevated and prolonged serum levels when taken with indomethacin, phenylbutazone, salicylates or sulfinpyrazone
  • Transient reduction in plasma concentrations of oestrogens and gestagens when administered with penicillins
  • Reduced absorption after intestinal sterilization with aminoglycosides
  • Prolonged prothrombin time/INR when combined with oral anticoagulants

Precautions for Pregnancy & Lactation

  • Crosses placenta
  • May be used throughout pregnancy with appropriate indication
  • Passes into breast milk in small amounts
  • May be used during lactation, but may cause diarrhoea and yeast colonization in the infant

Contraindications

  • Hypersensitivity to penicillins or any of the excipients
  • Severe gastrointestinal disorders accompanied by vomiting and diarrhea

  • Hypersensitivity reactions involving the skin
  • Serious allergic reactions
  • Anaphylactic or anaphylactoid reactions
  • Skin rashes or inflammation of mucous membranes
  • Gastrointestinal disturbances
  • Pseudomembranous colitis
  • Changes in blood picture
  • Herxheimer's reaction
  • Drug-induced aseptic meningitis
  • Transient discolouration of the teeth
  • Proliferation of resistant microorganisms

Precautions & Warnings

  • Possibility of cross-allergy between cephalosporins and penicillins
  • Consider potassium content for patients with heart diseases or serious electrolyte disturbances
  • Regular check of patient's condition for resistance proliferation and secondary infection
  • Consider sugar content for patients with diabetes mellitus
  • No impairment to drive or operate machinery, but predisposition to encephalopathy risk should be noted

Overdose Effects

  • Low toxicity
  • No acute toxicity for single multiple therapeutic dose
  • Risk of encephalopathy in case of overdose or renal impairment
  • No special measures required for overdose other than discontinuation of medication
  • Elimination through haemodialysis possible
