Indications
- Treatment of mild to moderately severe bacterial infections
- Infections of the ear, nose, and throat regions
- Infections of the lower respiratory tract
- Infections due to beta-hemolytic streptococci of group A
- Skin infections
- Lymphadenitis and lymphangitis
- Infections of the buccal cavity, gums, or jaws
- Prophylaxis of scarlet fever
- Prevention of recurrences of rheumatic fever
- Prophylaxis of infection after dental and oral surgical procedures
- Combination therapy with other antibiotics
Pharmacology
- Penicillin V is acid-stable
- Best absorbed from the upper part of the small intestine
- Convenient treatment of Gram-positive infections
- Advantage in resistance to gastric acid inactivation
Dosage
- Dosage determination based on sensitivity and severity of infection
- Adult dosage: 250-500 mg 6 hourly
- Children dosage: 125-250 mg 6 hourly
- Infants dosage: 62.5-125 mg 6 hourly
- Best taken with an empty stomach
Administration
- Best taken one hour before meals
- Tablets swallowed without chewing
- Syrup requires vigorous shaking before use
Interaction
- Reduction in absorption rate with concurrent food intake
- Potential drug interactions with methotrexate, probenecid, indomethacin, etc.
- Possible interference with laboratory and diagnostic tests
Contraindications
- Hypersensitivity to penicillins or excipients
- Severe gastrointestinal disorders with vomiting and diarrhea
Side Effects
- Hypersensitivity reactions involving the skin
- Serious allergic reactions
- Gastrointestinal disturbances
- Pseudomembranous colitis
- Changes in blood picture
- Occasional skin rashes and mucous membrane inflammation

Pregnancy & Lactation
- Crosses the placenta
- Passes into breast milk in small amounts
- Use during lactation may cause diarrhoea and yeast colonization
Precautions & Warnings
- Consider cross-allergy between cephalosporins and penicillins
- Consider potassium content in patients with heart diseases
- Check patient's condition at regular intervals
- Consider the sugar content in patients with diabetes mellitus
- No indication of impaired ability to drive or operate machinery
Overdose Effects
- Low toxicity with a broad therapeutic range
- Risk of encephalopathy in cases of overdose or renal impairment
- Elimination via haemodialysis
