Indications

  • Mild to moderately severe bacterial infections
  • Infections of the ear, nose, and throat
  • Bronchitis and pneumonia
  • Skin infections
  • Prevention of scarlet fever
  • Prophylaxis of infection after dental and oral surgical procedures
  • Combination therapy with other antibiotics

Pharmacology

  • Absorption from upper part of small intestine
  • Best absorbed in potassium salt form
  • Treatment of Gram-positive infections
  • Advantage over penicillin G in resistance to gastric acid inactivation

Dosage

  • Dosage determined by sensitivity of causative micro-organism and severity of infection
  • Adult dosage: 250-500 mg 6 hourly
  • Children dosage: 125-250 mg 6 hourly
  • Infants dosage: 62.5-125 mg 6 hourly

Administration

  • Best taken on empty stomach
  • Tablets swallowed without chewing with liquid
  • Syrup shaken vigorously before use

Interaction

  • Reduced absorption with concurrent food intake
  • Potential interactions with methotrexate, bacteriostatic antibiotics, probenecid, oral contraceptives, and oral anticoagulants

Contraindications

  • Hypersensitivity to penicillins or excipients
  • Severe gastrointestinal disorders accompanied by vomiting and diarrhea

Pregnancy & Lactation

  • Crosses placenta
  • Safe for use during pregnancy
  • Passes into breast milk in small amounts
  • Use during lactation may cause diarrhoea and yeast colonization in the infant

Precautions & Warnings

  • Consideration of cross-allergy with cephalosporins
  • Consideration of potassium content in patients with heart diseases or electrolyte disturbances
  • Regular checking of patient's condition for resistant microorganisms
  • Sugar content consideration in patients with diabetes mellitus
  • Encephalopathy risk

  • Low toxicity and broad therapeutic range
  • Risk of encephalopathy with beta-lactam antibiotics
