Medicine Details

Indications

  • Treatment of bacterial infections in ear, nose, and throat regions
  • Treatment of infections in lower respiratory tract
  • Treatment of infections due to beta-hemolytic streptococci of group A
  • Treatment of skin infections
  • Prophylaxis of scarlet fever
  • Prophylaxis of infection after dental and oral surgical procedures
  • Combination therapy with other appropriate antibiotics

Pharmacology

  • Absorption from upper part of small intestine
  • Best absorption with potassium salt form
  • Administration with meals
  • Resistance to inactivation by gastric acid

Dosage

  • Adult dosage: 250-500 mg 6 hourly
  • Children dosage: 125-250 mg 6 hourly
  • Infants dosage: 62.5-125 mg 6 hourly

Administration

  • Best taken on an empty stomach
  • Tablets swallowed without chewing
  • Syrup shaken vigorously before use

Interaction

  • Reduction in rate of absorption with concurrent food intake
  • Potential drug interactions with methotrexate, bacteriostatic chemotherapeutics, probenecid, oral anticoagulants, and more
  • Interference with laboratory and diagnostic tests

Contraindications

  • Hypersensitivity to penicillins or any of the excipients
  • Severe gastrointestinal disorders accompanied by vomiting and diarrhea

Side Effects

  • Hypersensitivity reactions involving the skin
  • Allergic reactions like drug fever, vasculitis, or interstitial nephritis
  • Anaphylactic or anaphylactoid reactions
  • Skin rashes or inflammation of mucous membranes

  • Herxheimer's reaction
  • Transient discolouration of the teeth

Pregnancy & Lactation

  • Crosses the placenta
  • Passes into breast milk in small amounts
  • May be used during pregnancy and lactation

Precautions & Warnings

  • Cross-allergy between cephalosporins and penicillins
  • Consideration of potassium content in patients with heart diseases or serious electrolyte disturbances
  • Risk of encephalopathy, particularly in case of overdose or renal impairment
  • Checking the patient's condition at regular intervals
  • Consideration of sugar content in patients with diabetes mellitus

Overdose Effects

  • Low toxicity with broad therapeutic range
  • Risk of encephalopathy, particularly in case of overdose or renal impairment
  • Elimination through haemodialysis
