Indications
- Treatment of mild to moderately severe bacterial infections
- Infections of the ear, nose, and throat regions
- Lower respiratory tract infections
- Infections due to beta-hemolytic streptococci
- Skin infections
- Prophylaxis of scarlet fever
- Prevention of recurrences of rheumatic fever
- Prophylaxis of infection after dental and oral surgical procedures
Pharmacology
- Absorption from the upper part of the small intestine
- Best absorbed as the potassium salt form
- Convenient means of treating Gram-positive infections
- Advantage over penicillin G in resistance to gastric acid inactivation
Dosage
- Dose determined by sensitivity of micro-organism and severity of infection
- Adult dosage: 250-500 mg 6 hourly
- Children dosage: 125-250 mg 6 hourly
- Infants dosage: 62.5-125 mg 6 hourly
Administration
- Best taken on an empty stomach
- Tablets are swallowed without chewing with liquid
- Syrup requires shaking before each use
Interaction
- Food intake reduces absorption rate
- Concomitant administration with penicillins may increase serum levels of methotrexate
- Possible disturbance of absorption of other orally administered drugs due to diarrhoea
- Attenuation or abolition of penicillin activity when combined with bacteriostatic chemotherapeutics or antibiotics
- Concurrent administration of probenecid inhibits renal excretion of penicillins
- May cause transient reduction in plasma concentrations of oestrogens and gestagens
Contraindications
- Hypersensitivity to penicillins or any of the excipients
- Severe gastrointestinal disorders accompanied by vomiting and diarrhea

- Hypersensitivity reactions involving the skin
- Serious allergic reactions
- Anaphylactic or anaphylactoid reactions
- Skin rashes or inflammation of mucous membranes
- Gastrointestinal disturbances
- Diarrhoea as a symptom of enterocolitis
- Possible blood picture changes
- Herxheimer's reaction
- Transient discolouration of the teeth
Pregnancy & Lactation
- Crosses placenta during pregnancy
- Passes into breast milk in small amounts
- May be used during lactation
Precautions & Warnings
- Possibility of cross-allergy between cephalosporins and penicillins
- Consideration of potassium content in patients with heart diseases or serious electrolyte disturbances
- Encephalopathy risk in case of overdose or renal impairment
- Regular condition checks for resistance micro-organisms proliferation
- Consideration of sugar content in patients with diabetes mellitus

- Risk of encephalopathy in cases of beta-lactam antibiotics administration
