Description
A tricyclic antidepressant with marked anticholinergic and sedative properties that prevents the reuptake of noradrenaline and serotonin at the nerve ending.
Mechanism of Action
Inhibit the membrane pump mechanism responsible for the re-uptake of transmitter amines, such as norepinephrine and serotonin, increasing their concentration at the synaptic clefts of the brain.
Dosage
- Depression: 75-200 mg daily
- Nocturnal Enuresis: 10-50 mg at night
- Prophylaxis of Migraine: 100 mg daily
- Tension Headache: 10-25 mg three times daily
Interactions
- Enhances the sedative effect of alcohol and opioid analgesics
- Increases the risk of ventricular arrhythmias when used with Moxifloxacin or Terfenadine
- Inhibition of metabolism with Disulfirum and Cimetidine
- Enhanced risk of postural hypotension when used with diuretics
Contraindications
- Myocardial infarction
- Arrythmias
- Heart block of any degree
- Mania
- Severe liver disease

Pregnancy & Lactation
Pregnancy Category C. Excreted into breast milk. Use during pregnancy only if the potential benefit justifies the potential risk to the fetus.
Precautions & Warnings
Use with caution in patients with epilepsy, glaucoma, urinary retention, cardiac disease, diabetes, pregnancy, hepatic impairment, thyroid disease, increased intra-ocular pressure and psychoses
