Description
Amtylin is a tricyclic antidepressant with marked anticholinergic and sedative properties. It prevents the reuptake of noradrenaline and serotonin at the nerve ending.
Indications
- Depressive Illness
- Nocturnal Enuresis in children
- Prophylaxis of Migraine
- Tension Headache
- Chronic Pain
Pharmacology
Amitriptyline inhibits the membrane pump mechanism responsible for the re-uptake of neurotransmitters, increasing their concentration at the synaptic clefts of the brain.
Dosage & Administration
- Depression: Initial dose of 75mg daily, increased gradually to 150-200mg
- Nocturnal Enuresis: Children 7-10 years: 10-20mg, 11-16 years: 25-50mg at night
- Prophylaxis of Migraine: 100mg daily
- Tension Headache: 10-25mg three times daily
Interaction
- Enhances sedative effect of alcohol and opioid analgesics
- Increases the risk of ventricular arrhythmias with Moxifloxacin or Terfenadine
Contraindications
- Myocardial infarction
- Arrhythmias
- Mania
- Severe liver disease
- Sedation may affect the ability to drive or operate machinery

Pregnancy & Lactation
