Description

Flucloxacillin is indicated for the treatment of infections due to Gram-positive organisms, including infections caused by penicillinase producing staphylococci. It is used for skin and soft tissue infections, respiratory tract infections, osteomyelitis, enteritis, endocarditis, urinary tract infection, meningitis, septicaemia, and as a prophylactic agent during major surgical procedures. It is active against Gram-positive organisms including penicillinase producing strains and has little activity against Gram-negative bacilli. The dosage varies for oral and parenteral administration, with specific recommendations for adults and children. Oral doses should be administered 1 hour before a meal. Concurrent use of Flac with certain medications may result in increased levels of Flac in the blood for a prolonged period. Flucloxacillin is contraindicated in penicillin hypersensitive patients. Common side effects include gastrointestinal disturbances, rashes, urticaria, purpura, fever, interstitial nephritis, hepatitis, and cholestatic jaundice. The US FDA Pregnancy Category of Flucloxacillin is B, and it is excreted in human milk. Caution should be exercised when Flucloxacillin is administered to a lactating mother. Caution should also be exercised in patients with evidence of hepatic dysfunction and those with an allergic diathesis. In severe renal failure, a reduction in dose or an extension of dose interval should be considered. The therapeutic class is penicillinase-resistant penicillins. Flac should be kept in a dry place away from light and heat, and out of the reach of children.

Pharmacology

  • Active against Gram-positive organisms
  • Inhibits formation of bacterial cell wall

Dosage

  • Oral administration for adults: 250 mg four times daily
  • Oral administration for children (2-10 years): Half of adult dose
  • Oral administration for children (Under 2 years): Quarter of adult dose
  • Parenteral administration for adults: 250 mg-1 g four times daily
  • Parenteral administration for children: Half of adult dose or a quarter of adult dose
  • Doses may be doubled in severe infections

Administration

  • Oral doses should be administered 1 hour before meal

Interaction

  • Concurrent use with certain medications may result in increased level of Flac in blood for prolonged period

Contraindications

  • Contraindicated in penicillin hypersensitive patients
