Indications
- Treatment of skin and soft tissue infections
- Treatment of respiratory tract infections
- Prophylactic use during major surgical procedures
- Treatment of osteomyelitis, enteritis, endocarditis, urinary tract infection, meningitis, septicaemia
Pharmacology
- Active against Gram-positive organisms
- Little activity against Gram-negative bacilli
- Inhibits formation of bacterial cell wall
- Bactericidal antibiotic
- Kills bacterial cell wall by interfering with peptidoglycan synthesis
Dosage
- Oral administration for adults: 250 mg four times daily
- Parenteral administration for adults: 250 mg-1 g four times daily
- Dosage may be doubled in severe infections
- Children dosage: 1/2 of adult dose for 2-10 years, 1/4 of adult dose for under 2 years
Administration
- Oral doses should be administered 1 hour before meal
Interaction
- Concurrent use may result in increased level of drug in blood
Contraindications
- Contraindicated in penicillin hypersensitive patients
Side Effects
- Common gastrointestinal side effects such as nausea, vomiting, diarrhoea, dyspepsia
- Rashes, urticaria, purpura, fever, interstitial nephritis, hepatitis, cholestatic jaundice
Pregnancy & Lactation
- US FDA Pregnancy Category: B
- Caution should be exercised when administered to a lactating mother
Precautions & Warnings
- Use with caution in patients with hepatic dysfunction
- Use caution in patients with an allergic diathesis
Use in Special Populations
- Reduce dose in severe renal failure (creatinine clearance <10 ml/min)

Reconstitution
- Has been used in other routes in conjunction with systemic therapy
- Administered by intraarticular and intrapleural injection
- Administered by nebuliser four times daily
