Tryptin is a tricyclic antidepressant with marked anticholinergic and sedative properties.

Inhibits the re-uptake of transmitter amines such as norepinephrine and serotonin
Increases concentration of neurotransmitters at synaptic clefts in the brain
Regulates mood by countering deficiencies in serotonin and norepinephrine neurotransmission

Depression: Initially 75 mg daily, increased gradually to 150-200 mg
Nocturnal Enuresis: Child 7-10 years: 10-20 mg, 11-16 years: 25-50 mg at night; max. period of treatment 3 months
Prophylaxis of Migraine: 100 mg daily
Tension Headache: 10-25 mg three times daily

Enhances sedative effect of alcohol and opioid analgesics
Increases risk of ventricular arrhythmias when used with Moxifloxacin or Terfenadine
Inhibited by Disulfiram and Cimetidine
Enhances risk of postural hypotension when used with diuretics

Anticholinergic: Excessive perspiration, dry mouth, blurred vision, hyperpyrexia, urinary retention
Cardiovascular reactions: Hypotension, syncope, postural hypotension, hypertension, tachycardia, palpitations
CNS and Neuromuscular: Confusional states, disorientation, delusions
Allergic: Skin rash, urticaria, photosensitization
Haematological: Bone-marrow depression
Gastrointestinal: Nausea, vomiting, diarrhoea, constipation
Endocrine: Testicular swelling, gynaecomastia, breast enlargement

Pregnancy Category C
Crosses the placenta
Excreted into breast milk
Use during pregnancy only if potential benefit justifies potential risk to fetus
Consideration should be made for nursing infants if continuing the drug

Use with caution in patients with a history of epilepsy, glaucoma, urinary retention, cardiac disease, diabetes, hepatic impairment, thyroid disease, increased intra-ocular pressure, and psychoses
