Indications

Pharmacology

  • Active against Gram-positive organisms
  • Little activity against Gram-negative bacilli
  • Inhibition of bacterial cell wall formation
  • Mechanism of action - interference with peptidoglycan synthesis
  • Bactericidal antibiotic
  • Useful against penicillinase-producing staphylococci

Dosage

  • Oral administration for adults: 250 mg four times daily
  • Double dosage in severe infections
  • Parenteral administration for adults: 250 mg-1 g four times daily
  • Parenteral administration for children: Half of the adult dose
  • Dosage may be doubled in severe infections

Administration

  • Oral doses to be administered 1 hour before meals

Interaction

  • Concurrent use may increase levels of Flucopen in blood

Contraindications

  • Contraindicated in penicillin hypersensitive patients

Side Effects

  • Common side effects: nausea, vomiting, diarrhea, dyspepsia
  • Other side effects: rashes, urticaria, purpura, fever, interstitial nephritis, hepatitis, cholestatic jaundice

Pregnancy & Lactation

  • FDA Pregnancy Category B
  • Caution in pregnant women
  • Caution in lactating mothers

Precautions & Warnings

  • Use with caution in patients with hepatic dysfunction
  • Use with caution in patients with allergic diathesis

Use in Special Populations

  • Caution in severe renal failure (creatinine clearance <10 ml/min)

  • Penicillinase-resistant penicillins

Reconstitution

  • Use in other routes in conjunction with systemic therapy
  • Intraarticular injection dosage: 250 mg to 500 mg daily
  • Intrapleural injection dosage: 250 mg daily
  • Inhalation by nebulizer dosage: 125 mg-250 mg four times daily
